Description
With this bridge grant, we went beyond the achievements of the previous My First AIRC Grant project by expanding the investigation to all human LH enzymes (the previous project focused on LH2), which convert collagen lysines into hydroxylysines, and by integrating the study of the further conversion of hydroxylysines into glucosyl-galactosyl-hydroxylysines. Using structural biology and biochemistry, we will determine the three-dimensional structures of these enzymes and characterize their function, with the ultimate goal of developing inhibitors capable of interfering with their activity and thereby reducing the impact of dysfunctions that can cause solid-tumour metastases. Developing inhibitors of LH-family enzymes represents an important step toward obtaining drugs capable of reducing the risk of, and potentially preventing, metastatic progression in multiple solid tumours. These basic-research investigations therefore have the potential for major future benefits for many cancer patients.
