To identify the physiological dimeric interface Scietti et al. introduced the Arg714Asn mutation in the PLOD3 recombinant protein. As verified by SDS-PAGE and enzymatic deglycosylation experiments this mutation added a novel glycosylation site at Asn714, in the middle of the tal-to-tail dimerization interface. This caused enzyme monomerization identifying the c-terminal interface as the physiological one.
