To identify the physiological dimeric interface Scietti et al. introduced the Thr672Asn mutation in the PLOD3 recombinant protein. As verified by SDS-PAGE and enzymatic deglycosylation experiments this mutation added a novel glycosylation site at Asn672. This caused enzyme monomerization identifying the c-terminal interface as the physiological one. |
